Rapid synthesis of pomalidomide-conjugates in minutes via temperature elevation and delayed feeding

Abstract

The synthesis of pomalidomide-conjugates, crucial intermediates for developing PROTACs, suffers from prolonged reaction times and extensive byproduct formation. Herein, we present a methodology that overcomes these limitations, achieving complete reaction within minutes while minimizing side products. By shortening the reaction timeframes, our method will facilitate the exploration of novel pomalidomide-based PROTACs.
